Tough choice since most movies and games make my tears fall out when it comes to this.
The one from childhood was "Hey You Pikachu" since I couldn't separate from the chu that hung out with me for weeks. But it was awfully sweet that he came back. ^_^
I also loved how sweet "UP" was with how it all ended. Carl being the stand-in father for Russel, Kevin reuniting with her babies, and the HOUSE landing in the right spot was really heart touching.
But the reason why I loved Astro Boy was that his movie was playing in the hospital I was in when I suffered an infection. I just caught the movie at the moment when the boy robot was in the parade but seeing just an (seemingly) innocent kid forced into battle was just SAD. His cute remark about the machine guns brought a bit of joy, but when he took his life to save the city, I just couldn't hold back the tears. Even when Astro was revived I was still crying at the end. Cheesy it is, but thank goodness the boy robot made things better in there.

Hmm, trying to think of one was surprisingly hard, for music especially, seeing as just trying to think of one example out of the ridiculous amount of music i've got seemed impossible. But i'd say the one thats left the biggest impression on me is Empire Ants by Gorillaz. Particuly, the live version they did at this years Glastonbury festival, which i think is slightly better then the album version;

It's a combination of the melodies and chords throughout the song (and the beautiful synths and strings they're played on), mixed with the lyrics sung by the guest vocalist; "Little memories, marching on; your little feet, working the machine; will it spin, will it soar; my little dream, working the machine". I honestly can't explain why, but something about the lyrics grabbed me from the get go. That, and i admit, i love the section with the climbing string notes (4:45 - 4:55).

Astro: ¬__¬ Would it KILL YOU to be optomistic and posetive for once in your life?

The ending to The Walking Dead video game. It ripped the heart out from my chest and made the father/son scene between Astro and Tenma at the end of 2003 anime look like fanfiction written by a duck!

For something Astro Boy related I think one of the saddest moments is in the special The Secret of Atom's Birth when there's a flash back to Tobio's funeral (oh dear god). Pretty much that whole special was enough to be moving.
And I guess for a non-Tezuka example for me comes from Pinocchio. 'When You Wish Upon A Star' never fails in getting me to tear up.
The ending of Edward Scissorhands was very sad too.

In terms of saddest, I think that the death that happens in Episode 12 of Now and Then, Here and There takes precedence in my mind. I won't spoil who dies for whoever wants to watch that series. I am not one to cry often during movies, anime, manga, video games, TV shows, or things of that sort, but that death is the only time I have ever come very close to crying during an anime.
I don't know what would be the sweetest for me, but I saw Wreck-It Ralph last night and that movie had some sweet moments. It tugs at your heartstrings at certain points.

The first time any media made me tear up was the ending of the door stopper novel Les Miserables. Really, the reason I love the book so much was because the last two/three hundred pages were unbelievable and the ending was so moving.
The last Dark Tower book made me almost cry three separate times.
Several moments in Zatch Bell made me get a little teary eyed, but nothing has (so far) come close to the impact Les Mis had on me.
And, of course, when Atom confronts Tenma at the end of the '03 series.
